In its supplied form epitalon is a white to off-white powder, usually lyophilized and often hygroscopic. It dissolves readily in water and in aqueous buffers, and it is commonly handled as the acetate or trifluoroacetate salt to improve stability during purification and drying. The amide backbone is labile in aqueous media, so solutions are less durable than the dry solid. Handling notes in the chemical literature therefore emphasize keeping the powder dry, cool and shielded from light until it is dissolved.
Epitalon is a synthetic tetrapeptide whose sequence is alanine-glutamate-aspartate-glycine, written in single-letter code as AEDG. The four residues are joined by three peptide bonds, giving a linear backbone with no branching and no disulfide bridges. Its calculated molecular mass for the free form is approximately 390.3 daltons, a figure that rises when the compound is supplied as an acetate or trifluoroacetate salt. Because the chain is short, the molecule is defined entirely by its residue order rather than by any folded three-dimensional structure.
The compound is described in the literature as a derivative of epithalamin, a preparation obtained from bovine pineal tissue. Work on this peptide family was carried out mainly by a research group in Saint Petersburg beginning in the 1980s, and the substance was later registered for clinical use in Russia under the name Epitalon. Outside that region it is generally treated as a research chemical rather than an approved medicine. Statements about its biological activity rest on a relatively small number of studies, and independent replication remains limited.
No national medicines regulator has approved epitalon as a therapeutic product. It is generally distributed as a research chemical, and in some jurisdictions selling peptides for human consumption without approval is restricted or prohibited. Certificates of analysis accompanying commercial material vary in which tests are performed, and independent verification of identity and purity is uncommon. Statements about anti-ageing or disease-prevention benefits on vendor pages are marketing claims rather than regulatory findings, a distinction that shapes how the compound is discussed in scientific and popular sources alike.
The most frequently cited laboratory finding is that AEDG increased telomerase activity and extended telomere length in cultured human somatic cells. That work used fetal fibroblast strains and reported changes in enzyme activity alongside altered division counts. Replication by unrelated groups has been limited, and the published record is largely a single-laboratory series rather than a multi-centre programme. The result supports a hypothesis about peptide influence on gene expression in cell culture; it does not by itself establish an effect on telomere length in living animals or in people.
Animal and clinical reports appear mainly in Russian-language journals from the 1990s and 2000s, covering endpoints such as melatonin rhythm, lifespan in aged rodents, and retinal function. Many of these papers involve small groups, lack blinding or placebo comparison, and are difficult to retrieve through indexed databases. Review articles published in English generally summarise the claims without reanalysing the underlying data. Because no large randomised trial exists, the clinical importance of these reported effects stays unresolved and is properly described as an open question.

Epitalon is a synthetic tetrapeptide with the residue sequence alanine-glutamate-aspartate-glycine, commonly abbreviated AEDG. Its monoisotopic mass is approximately 390.35 daltons, and it is usually supplied as a lyophilised trifluoroacetate or acetate salt. The compound was derived from a pineal gland extract called epithalamin, a heterogeneous preparation investigated in the former Soviet Union. Researchers associated with the Saint Petersburg Institute of Bioregulation and Gerontology described the tetrapeptide as a constituent fragment of that extract. Commercial material is offered as a laboratory reagent rather than as a finished pharmaceutical product.
Literature searches for this compound must account for several spelling variants. Indexing databases contain epitalon, epithalon, epithalone, and AEDG, and relevant records are scattered across Russian-language and English-language journals that do not consistently cross-cite. Early publications describe the parent extract as a mixture of many peptides, whereas later work addresses the single synthetic tetrapeptide. That shift in nomenclature complicates comparison between studies, because extract data and tetrapeptide data are sometimes cited interchangeably. A search strategy omitting the alternate spellings will return an incomplete set of references.
Reported biological findings come mainly from cell culture and rodent experiments. Those studies describe changes in telomerase catalytic subunit expression, melatonin rhythm amplitude, and antioxidant enzyme activity after peptide exposure. Human data are sparse and consist of small trials with limited blinding and inconsistent endpoints. The proposed mechanisms remain hypotheses rather than established facts, and there is no consensus on whether effects observed in animals carry over to people. Independent replication outside the original research groups is limited, which is a recognised gap in the published literature.
Identity is normally established by reversed-phase high-performance liquid chromatography combined with mass spectrometry, a pairing that separates components and confirms molecular mass at once. Purity is quoted as a percentage from the chromatogram, and figures above ninety-five percent are a frequent commercial specification. Amino acid analysis or sequencing supplies further confirmation when required. Because many short peptides behave similarly under chromatography, retention time alone does not establish sequence; the mass measurement is what separates one tetrapeptide from another, and certificates should report both.
Short peptides such as AEDG are normally supplied as a freeze-dried solid and are kept dry, cold, and shielded from light. Holding at minus twenty degrees Celsius is common for the long term, while a refrigerator suffices for brief periods before use. The material takes up moisture to some degree, so containers should stay sealed and be allowed to reach room temperature before opening, which limits condensation on the contents. Repeated warming and cooling of a single container is discouraged because it admits water and can lower the amount of intact peptide.
The four residues give epitalon a molecular formula of C14H22N4O9 and a molecular weight near 390.35 daltons. The presence of two acidic residues, glutamate and aspartate, makes the free peptide strongly acidic, while the alanine and glycine ends provide neutral, nonpolar character. This combination produces a molecule with substantial water solubility. Because there are no cysteine, methionine, or tryptophan residues, the peptide lacks the most common oxidation-sensitive side chains, which simplifies handling compared with many longer peptides.
The proposed relationship between epitalon and pineal function is a central part of its background. Khavinson's group reported that short peptides corresponding to regions of larger pineal proteins could influence gene expression in cells. Epitalon was framed as a synthetic analogue of an active fragment rather than a direct isolation product. Whether the tetrapeptide reproduces the effects of the parent extract is an open question, because comparative studies are limited and the parent extract itself is not a single defined substance.
Epitalon emerged from research conducted in Saint Petersburg by Vladimir Khavinson and colleagues, who studied short peptides as potential regulators of aging. The work built on epithalamin, a pineal gland extract reported to influence neuroendocrine function. Epitalon was designed as a synthetic counterpart with a defined sequence, allowing reproducible experiments that extracts could not support. Early publications described effects on melatonin rhythms and lifespan in animal models. These findings circulated mainly in Russian-language journals during the 1990s, which limited their visibility among English-speaking researchers.
The most widely cited claim is that epitalon activates telomerase and thereby extends telomere length. Supporting evidence comes largely from cultured human cells, where treatment was associated with increased telomerase activity and delayed replicative senescence. Telomerase activation is a biologically consequential effect, since the enzyme is largely silenced in most somatic cells. However, the route by which a short peptide would reach and act on the enzyme's regulatory machinery has not been established. Independent replication in human trials is scarce, so the link between cell-culture observations and whole-body aging remains an open question.

=== Drug development === LC–MS is frequently used in drug development because it allows quick molecular weight confirmation and structure identification. These features speed up the process of generating, testing, and validating a discovery starting from a vast array of products with potential application. LC–MS applications for drug development are highly automated methods used for peptide mapping, glycoprotein mapping, lipidomics, natural products dereplication, bioaffinity screening, in vivo drug screening, metabolic stability screening, metabolite identification, impurity identification, quantitative bioanalysis, and quality control.
In order to locate Razzouki, we needed the same techniques that were used 30 years ago to track down Pablo Escobar, because Razzouki applied the same routines in order to stay under the radar and escape every surveillance by the Colombian authorities. Razzouki did not use any technology or social media for eight years, to the extent that he did not update his family in the Netherlands of any news. Our investigators tracked him down by using intelligence planes that regularly flew over the city of Medellín with the aim of locating him. The last time the Elite Colombian Police Unit intervened in such a case, was during the operation carried out on 2 December 1993 to track down and shoot Pablo Escobar. Razzouki is suspected of drug trafficking, manufacture and possession of illegal substances, homicide, and terrorism. He had changed his physical appearance with the help of cosmetic surgery in order to look older. Razzouki belonged to a European criminal organisation of the Moroccan Mafia and was at the head of a group of traffickers from the Clan del Golfo cartel who were involved in the international export of cocaine, especially to Spain and Portugal. The Iranian government allegedly protected Taghi as he worked together with the Iranian secret service, to assassinate enemies of the Iranian state who were hiding out on European soil. On 16 December 2019, Taghi was arrested in his mansion in Dubai after a manhunt by the Moroccan and European authorities, and three days later he was deported to the Netherlands.
RPAs allow for the determination of the presence of altered proteins or other agents that may be the result of disease. Specifically, post-translational modifications, which are typically altered as a result of disease can be detected using RPAs.
== Structure == Fibronectin exists as a protein dimer, consisting of two nearly identical polypeptide chains linked by a pair of C-terminal disulfide bonds. Each fibronectin subunit has a molecular weight of approximately 230–275 kDa and contains multiple copies of three types of modules: type I, II, and III. All three modules are composed of two anti-parallel β-sheets resulting in a beta-sandwich; however, type I and type II are stabilized by intra-chain disulfide bonds, while type III modules do not contain any disulfide bonds. The absence of disulfide bonds in type III modules allows them to partially unfold under applied force. Three regions of variable splicing occur along the length of the fibronectin protomer. One or both of the "extra" type III modules (EIIIA and EIIIB) may be present in cellular fibronectin, but they are never present in plasma fibronectin. A "variable" V-region exists between III14–15 (the 14th and 15th type III module). The V-region structure is different from the type I, II, and III modules, and its presence and length may vary. The V-region contains the binding site for α4β1 integrins. It is present in most cellular fibronectin, but only one of the two subunits in a plasma fibronectin dimer contains a V-region sequence. The modules are arranged into several functional and protein-binding domains along the length of a fibronectin monomer. There are four fibronectin-binding domains, allowing fibronectin to associate with other fibronectin molecules.

=== Depression === A comparative meta-analysis of 21 major antidepressants found that venlafaxine, agomelatine, amitriptyline, escitalopram, mirtazapine, paroxetine, and vortioxetine were more effective than other antidepressants, although the quality of many comparisons was assessed as low or very low. Open-label evidence also suggests potential benefit in treatment-resistant cases: in a multicenter Canadian study of adults with inadequate response to prior antidepressants (n=159), 58% achieved response and 28% remission after 8 weeks of venlafaxine (mean 260 mg/day); tolerability was generally acceptable. Venlafaxine was similar in efficacy to the atypical antidepressant bupropion; however, the remission rate was lower for venlafaxine. In a double-blind study, patients who did not respond to an SSRI were switched to either venlafaxine or another SSRI (citalopram); similar improvement was observed in both groups. Studies have not established its efficacy for use in pediatric populations. In children and adolescents with depression, venlafaxine increases the risk of suicidal thoughts or attempts. Higher doses (e.g., 225 mg and 375 mg per day) of venlafaxine are more effective than lower doses (e.g., 75 mg per day) but also cause more side effects. Studies have shown that the extended-release is superior to the immediate-release form of venlafaxine. A 2017 meta-analysis has shown that the efficacy of venlafaxine is not correlated with baseline severity of depression.
Duterte was elected unopposed for his seventh term in 2013. In November of that year, Davao City sent rescue and medical teams to Tacloban to give aid to the victims of Typhoon Haiyan (locally known in the country as Typhoon Yolanda). Financial assistance was also given to Bohol and Cebu for earthquake victims. Duterte also passed Davao City's Women Development Code, which aims "to uphold the rights of women and the belief in their worth and dignity as human beings". Duterte banned swimsuit competitions in beauty pageants in Davao City. He gained prominence for supporting the first-ever Gawad Kalinga Village inside a jail facility in Davao City. It is a home-type jail with ten cottages built inside the compound, which now serve as homes for female inmates.

=== Breast cancer === Breast cell proliferation has been found to be significantly increased by the combination of an oral estrogen plus cyclic medroxyprogesterone acetate in postmenopausal women but not by the combination of transdermal estradiol plus oral progesterone. Studies of topical estradiol and progesterone applied to the breasts for 2 weeks have been found to result in highly pharmacological local levels of estradiol and progesterone. These studies have assessed breast proliferation markers and have found increased proliferation with estradiol alone, decreased proliferation with progesterone, and no change in proliferation with estradiol and progesterone combined. In the Postmenopausal Estrogen/Progestin Interventions (PEPI) trial, the combination of estrogen and cyclic oral progesterone resulted in a higher mammographic breast density than estrogen alone (3.1% vs. 0.9%) but a non-significantly lower breast density than the combination of estrogen and cyclic or continuous medroxyprogesterone acetate (3.1% vs. 4.4–4.6%). Higher breast density is a strong known risk factor for breast cancer. Other studies have had mixed findings however. A 2018 systematic review reported that breast density with an estrogen plus oral progesterone was significantly increased in three studies and unchanged in two studies. Changes in breast density with progesterone appear to be less than with the compared progestins.

The peptide contains four residues in the order alanine, glutamate, aspartate and glycine, abbreviated AEDG. The chain is linear and held together by three peptide bonds. Its calculated mass for the uncharged free form is about 390.3 daltons.
Epithalamin refers to a crude preparation derived from bovine pineal tissue, which contains many components. Epitalon is a single synthetic tetrapeptide identified from that material and produced by chemical synthesis. The two names are sometimes confused because early reports treated the synthetic peptide as an active fragment of the extract.
It has been registered for clinical use in Russia, where it is associated with a small family of short peptides. In most other jurisdictions it is not an approved medicine and is traded as a research chemical. Regulatory status varies by country and changes over time.
Small studies with human participants have been reported, chiefly in Russian-language journals, but they are limited in size and design. No large randomised controlled trial with published results is available.
