The short version of lyophilization fits in a sentence. The long version — which is the one that helps — is below.

Regulatory status varies by country and is not harmonized. Epitalon is not an approved drug in major Western jurisdictions. In some countries it is sold as a research chemical, and in others it has appeared in products marketed for other categories. This inconsistent status means that purity, labeling accuracy, and documentation differ widely between suppliers, and verification of identity and purity through independent analysis is the usual way buyers assess a given lot.
Epitalon appears in the literature primarily through a small number of research groups, most of them associated with the St. Petersburg Institute of Bioregulation and Gerontology. Publications describe cell culture experiments, animal studies, and a limited number of small human studies. Independent replication outside this network is sparse. As a result, reviews of the topic usually separate descriptive reports of observed effects from the question of whether those effects generalize, and they note the difficulty of comparing studies that use different preparations and endpoints.
Analytical characterization of epitalon relies on standard peptide methods. Reverse-phase high-performance liquid chromatography is used to assess purity, and mass spectrometry confirms identity by checking the observed mass against the expected value near 390 daltons. Amino acid analysis can verify composition. Because the peptide is short and hydrophilic, it elutes early on many reverse-phase columns, so method development often requires ion-pairing reagents to achieve adequate retention and resolution from related impurities.
Literature searches for this compound must account for several spelling variants. Indexing databases contain epitalon, epithalon, epithalone, and AEDG, and relevant records are scattered across Russian-language and English-language journals that do not consistently cross-cite. Early publications describe the parent extract as a mixture of many peptides, whereas later work addresses the single synthetic tetrapeptide. That shift in nomenclature complicates comparison between studies, because extract data and tetrapeptide data are sometimes cited interchangeably. A search strategy omitting the alternate spellings will return an incomplete set of references.
Reported biological findings come mainly from cell culture and rodent experiments. Those studies describe changes in telomerase catalytic subunit expression, melatonin rhythm amplitude, and antioxidant enzyme activity after peptide exposure. Human data are sparse and consist of small trials with limited blinding and inconsistent endpoints. The proposed mechanisms remain hypotheses rather than established facts, and there is no consensus on whether effects observed in animals carry over to people. Independent replication outside the original research groups is limited, which is a recognised gap in the published literature.

Identity and purity of epitalon samples are normally established by reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres, combined with mass spectrometry. The mass spectrum confirms the expected molecular ion and can reveal truncated or oxidised by-products. Amino acid analysis after acid hydrolysis verifies that the four residues are present in the expected ratio. Certificates typically report a purity figure taken from chromatographic peak area, expressed as a percentage of total integrated signal.
Lyophilised epitalon is generally held at minus twenty degrees Celsius in a sealed container kept dry and dark. Cooler conditions are sometimes recommended for long-term archives. The solid takes up moisture readily enough that repeated opening of a vial introduces water, so dividing a batch into smaller portions before storage lowers degradation risk. Aqueous solutions are less durable than the dry powder and are usually prepared shortly before use, then kept cold and shielded from light to slow hydrolysis and oxidation.
Verification of research-grade material involves comparing a supplier chromatogram against an in-house reference, checking the observed mass against the calculated value, and where possible confirming residue order by tandem mass spectrometry or enzymatic peptide mapping. Purity claims should be read alongside the method used to obtain them, because detection wavelength and integration settings alter the result. Batch-specific data, rather than a generic grade statement, is the informative part of a certificate.
Epitalon is a synthetic tetrapeptide with the sequence alanine-glutamate-aspartate-glycine, commonly abbreviated AEDG. It was designed at the St. Petersburg Institute of Bioregulation and Gerontology as a short, chemically defined analogue of epithalamin, a fraction obtained from bovine pineal tissue. Small peptide bioregulators of this type formed a long-running line of work there from the 1980s onward. Because the molecule is produced by solid-phase synthesis rather than extraction, its composition is exact and its purity can be measured directly.
The most frequently cited proposed action is induction of telomerase, the enzyme that adds repeat sequences to chromosome ends. Cell-culture work from the originating group reported higher telomerase reverse transcriptase expression and measurable telomere elongation in human somatic cells after exposure. How a four-residue peptide would reach nuclear gene regulation is not established, and no cell-surface receptor or uptake route has been identified. Additional reports describe changes in melatonin secretion, antioxidant enzyme activity and lipid peroxidation in aged animals, but these findings remain mechanistically unconnected to the telomerase observation.
Published evidence comes mainly from Russian-language journals and from a single research group, with small sample sizes and limited independent replication. A few laboratories outside that group have examined related peptides and reported weaker or absent telomerase effects, so the central claim is best described as contested rather than settled. Rodent studies report modest changes in some ageing markers and in survival, but designs vary and control conditions are often sparse. No large randomised trial in humans has been published, and long-term safety data in healthy populations are correspondingly thin.
Reversed-phase high-performance liquid chromatography is the standard approach for assessing purity, usually with ultraviolet detection near 214 nm, where the peptide bond absorbs. Mass spectrometry, most often with electrospray ionization, confirms the molecular mass and helps reveal truncation or deletion byproducts. Amino acid analysis can verify composition, and counterion content is sometimes measured because peptides purified with trifluoroacetic acid retain variable amounts of that salt. Purity figures reported without a stated method and detection wavelength are difficult to interpret.
Material sold for research use varies widely in documented quality. A useful verification package includes a certificate of analysis that states peptide content rather than only net weight, the chromatographic method and column used, and a mass spectrum consistent with the expected mass. Independent testing by a third-party laboratory is occasionally reported. Statements of identity resting only on a supplier label provide little assurance, and the gap between nominal mass and actual peptide content can be substantial once counterions and residual water are counted.
== Effects on animals == A 10 g vicine /kg diet in laying hens led to reduced feed intake, egg weight, haemoglobin levels and fertility and increased liver weights, liver glutathione levels and plasma lipid levels. A diet with comparable levels of vicine per kg in pigs showed only small effects on protein and energy digestibility. In another study, laying and broiler hens were fed grains that were soaked for different periods of time, which partly or totally removed vicine. Hens that had had grains with vicine still in them showed a significant decrease in corpuscular haemoglobin, while the others did not. An in vivo study in rats showed that oral administration of vicine resulted in only small reductions in glutathione concentrations and no mortality. Intraperitoneal administration however, led to a rapid decrease in glutathione followed by death because of anoxia.
Because peptide chains have a directionality conferred by their N-terminus and C-terminus, β-strands too can be said to be directional. They are usually represented in protein topology diagrams by an arrow pointing toward the C-terminus. Adjacent β-strands can form hydrogen bonds in antiparallel, parallel, or mixed arrangements. In an antiparallel arrangement, the successive β-strands alternate directions so that the N-terminus of one strand is adjacent to the C-terminus of the next. This is the arrangement that produces the strongest inter-strand stability because it allows the inter-strand hydrogen bonds between carbonyls and amines to be planar, which is their preferred orientation. The peptide backbone dihedral angles (φ, ψ) are about (–140°, 135°) in antiparallel sheets. In this case, if two atoms Cαi and Cαj are adjacent in two hydrogen-bonded β-strands, then they form two mutual backbone hydrogen bonds to each other's flanking peptide groups; this is known as a close pair of hydrogen bonds. In a parallel arrangement, all of the N-termini of successive strands are oriented in the same direction; this orientation may be slightly less stable because it introduces nonplanarity in the inter-strand hydrogen bonding pattern. The dihedral angles (φ, ψ) are about (–120°, 115°) in parallel sheets.
The treatment of slaves in the United States varied widely depending on conditions, times, and places. The power relationships of slavery corrupted many whites who had authority over slaves, with children showing their own cruelty. Masters and overseers resorted to physical punishments to impose their wills. Slaves were punished by whipping, shackling, hanging, beating, burning, mutilation, branding and imprisonment. Punishment was most often meted out in response to disobedience or perceived infractions, but sometimes abuse was carried out to re-assert the dominance of the master or overseer of the slave. Treatment was usually harsher on large plantations, which were often managed by overseers and owned by absentee slaveholders. William Wells Brown, who escaped to freedom, reported that on one plantation, slave men were required to pick 80 pounds (36 kg) of cotton per day, while women were required to pick 70 pounds (32 kg) per day; if any slave failed in their quota, they were subject to whip lashes for each pound they were short. The whipping post stood next to the cotton scales. A New York man who attended a slave auction in the mid-19th century reported that at least three-quarters of the male slaves he saw at sale had scars on their backs from whipping. By contrast, small slave-owning families had closer relationships between the owners and slaves; this sometimes resulted in a more humane environment but was not a given.
On 20 January 2001 the Indian Space Research Organisation (ISRO) conducted a balloon flight from Hyderabad, India to collect stratospheric dust from a height of 41 km (135,000 ft) with a view to testing for the presence of living cells. The collaborators on this project included a team of UK scientists led by Wickramasinghe. In a paper presented at a SPIE conference in San Diego in 2002 the detection of evidence for viable microorganisms from 41 km above the Earth's surface was presented. However, the experiment did not present evidence as to whether the findings are incoming microbes from space rather than microbes carried up to 41 km from the surface of the Earth. In 2005 the ISRO group carried out a second stratospheric sampling experiment from 41 km altitude and reported the isolation of three new species of bacteria including one that they named Janibacter hoylei sp.nov. in honour of Fred Hoyle. However, these facts do not prove that bacteria on Earth originated in the cosmic environment. Samplings of the stratosphere have also been carried out by Yang et al. (2005, 2009). During the experiment strains of highly radiation-resistant Deinococcus bacterium were detected at heights up to 35 km. Nevertheless, these authors have abstained from linking these discoveries to panspermia. Wickramasinghe was also involved in coordinating analyses of the red rain in Kerala in collaborations with Godfrey Louis.

The resulting viral mRNA looks is identical to host mRNA, allowing endogenous cellular machinery to carry out processing and nuclear export. The de-capped host mRNAs are targeted degradation, which lead to the downregulation of cellular mRNA. Influenza RdRp also interacts with the cell Polymerase II (Pol II) C terminal domain, which potentially promotes viral transcription by changing the conformation of the RdRp. Additionally, by reducing Pol II abundance, influenza can begin to shut off critical host transcription. Cap snatching is not used during replication. Instead, the RdRp performs a "prime and realign" step ensure that the genome is fully copied. In this mechanism, the RdRp sets down a primer internally, then the vRNA is realigned to continue replication. Influenza's PB2 cap-binding domain has a unique fold, but it uses aromatic stacking to execute m7G cap-binding similar to other cap-binding proteins. PA is a member of the PD(D/E)XK nuclease family, which uses divalent metal ions to cleave nucleic acid. However, it has a peculiar active site histidine residue which ligates the Mn2+ ion used for cleavage.
=== Hemolytic crisis === A hemolytic crisis, or hyperhemolytic crisis, is characterized by an accelerated rate of red blood cell destruction leading to anemia, jaundice, and reticulocytosis. Hemolytic crises are a major concern with sickle-cell disease and G6PD deficiency.
== Legacy == Half-Life 2: Episode Three was scheduled for release by Christmas 2007. It was canceled after Valve abandoned episodic development and began developing a new game engine, Source 2. After canceling several further Half-Life games, Valve released Half-Life: Alyx in 2020. In November 2024, Valve delisted Episode One and Two from the Steam Store and incorporated them into Half-Life 2. In reference to the Episode Two achievement "Little Rocket Man", which requires the player to carry a garden gnome from the start to the end of the game and place it into a rocket before it launches into orbit, Newell partnered with Wētā Workshop and Rocket Lab to create and launch a garden gnome on their "Return To Sender" space mission. The mission launched on November 20, 2020, from Mahia Launch Complex, New Zealand, as a mass simulator.

=== Outcomes === Treatment with docetaxel increases survival time in people with certain types of cancer. While some clinical trials show median survival times to be increased by approximately only three months, the range of survival time is large. Many people survive beyond five years with treatment from docetaxel, however it is difficult to attribute these findings directly to treatment with docetaxel. Improved median survival time and response indicates that docetaxel slows metastatic cancer progression and can lead to disease-free survival. Conjunctive treatment of prednisone with docetaxel has been shown to lead to improved survival rate as well as improved quality of life and reduction of pain compared with treatments with mitoxantrone. As well as inhibiting mitosis, the presence of docetaxel has been found to lead to the phosphorylation of the oncoprotein bcl-2, which leads to apoptosis of cancer cells that had previously blocked the apoptotic inducing mechanism, leading to tumour regression. Enhanced effects of radiation therapy when combined with docetaxel has been observed in mice. Docetaxel has also been found to have greater cellular uptake and is retained longer intracellularly than paclitaxel allowing docetaxel treatment to be effective with a smaller dose, leading to fewer and less severe adverse effects.
==== Obesity and osteoarthritis ==== Osteoarthritis and obesity are closely linked. Obesity is one of the most important preventable factors for the development of osteoarthritis. Originally, the relationship between osteoarthritis and obesity was considered to be exclusively biomechanically based, according to which the excess weight caused the joint to become worn down more quickly. However, today we recognise that there is also a metabolic component which explains why obesity is a risk factor for osteoarthritis, not only for weight-bearing joints (for example, the knees), but also for joints that do not bear weight (for example, the hands). Consequently, it has been shown that decreasing body fat lessens osteoarthritis to a greater extent than weight loss per se. This metabolic component related with the release of systemic factors, of a pro-inflammatory nature, by the adipose tissues, which frequently are critically associated with the development of osteoarthritis. Thus, the deregulated production of adipokines and inflammatory mediators, hyperlipidaemia, and the increase of systemic oxidative stress are conditions frequently associated with obesity, which can favour joint degeneration. Furthermore, many regulation factors have been implicated in the development, maintenance, and function, both of adipose tissues, as well as of the cartilage and other joint tissues. Alterations in these factors can be the additional link between obesity and osteoarthritis.

Identity is normally confirmed by mass spectrometry, which checks the measured mass against the expected value near 390 daltons. Reverse-phase high-performance liquid chromatography is used alongside it to assess purity. Amino acid analysis can provide additional composition data.
The powder is generally kept refrigerated or frozen, protected from light and moisture. Vials should reach room temperature before opening to prevent condensation. Reconstituted solutions are usually stored cold and used within a limited window because dilute solutions can degrade or support microbial growth.
Epitalon is not an approved drug in major Western regulatory jurisdictions. Its legal status differs between countries, and it is often distributed as a research chemical. This means product documentation and purity vary considerably between suppliers.
It is based on epithalamin, a peptide-rich preparation obtained from pineal gland tissue and studied in the former Soviet Union. Researchers later identified the tetrapeptide AEDG as a constituent fragment of that material. Modern supplies are produced by chemical synthesis rather than tissue extraction.
